New breakthrough by Prof. Lu, Xincheng’s research team published in Nature Communications

Prof. Lu, Xincheng, a researcher from the genome research institute of Wenzhou Medical University, published in Nature Communications (impact factor 10.02, part of the Nature Publishing Group) his latest paper, RSP02-LGR5 signaling has tumor-suppressive activity in colorectal cancer, which reveals the new role RSP02 plays in the genesis and development of colorectal cancer, and a novel mechanism via which it affects Wnt signal route, through its interaction with LGR5. The first authors of the paper are Wu Changjie, Qiu Sunquan, both of whom post-graduates and Lu Liting, a Lab technician of Prof. Lu Xincheng’s research team, and Prof. Lu’s byline appears as an interdependent correspondent. All the research works of this discovery were conducted in WMU, which demonstrates that the research capability of Prof. Lu’s research team has reached international excellence. 

 

RSP02 , which belongs to the R-spondin family, is a type of secretive protein with the capacity to trigger Wnt signal. It plays a vital role in many processes such as cell reproduction, embryo development and bone formation. In light of the fact that many colorectal cancer cases start in concomitance with an abnormal activation of Wnt signal, RSP02 is suspected to be a potential proto-oncogene for colorectal cancer. The research of Prof. Lu’s team observed that RSP02 actually functions as an anti-oncogene, with inhibitive effect on the genesis of colorectal cancer. The paper confirms the inhibitive mechanism, in which RSP02 activates the high expression of LGR5, and through the subsequent interaction to form the suppression of the Wnt/βcatenin feedback. The research on its molecular mechanism also found out that RSP02-LGR5 signal impairs Wnt signal and prompts the catabolization of LRP6 via stabilizing the ZNRF3 protein on the cell membrane. The discovery of Prof. Lu’s team reveals a previously unfamiliar type of Wnt feedback mechanism, which is not only of great significance to the study of Wnt signal route, but also provides theoretic foundation to the development of RSP02 as a new target point for the treatment of human colorectal cancer. The project was sponsored by the National Natural Science Foundation, and the Starting Fund for Wenzhou Medical University Foreign Talents.

 

Prof. Lu, a research team leader who came to Wenzhou Medical University from the USA in 2009, was one of the founding members of the WMU Genome Medicine Institute. Over the years, his team has published 13 SCI papers in premium international science journals including Nature Communications, J Hepatology, Nat Cell Biology, Cell Cycle, J Mol Cell Biol, among which 5 were published with WMU as either first author or correspondent. These papers has amassed in total 30 impact factors. Prof. Lu’s research team has also been granted with 3 sponsorships from the National Natural Science Foundations, 2 Merit-Based Supporting Funds. Prof. Lu also servs as one of the key academics in the National 973 Projects.
